《幕后真凶》是一部以缉毒警察为题材的案件悬疑剧，剧情跨越了20多年，讲述了两代缉毒警察的故事。三十多年前，在松江土地上发生了一起生物制剂非法出境案，随着此案的发生，缉毒刑警计春生失踪，卧底警察楚江河生死不明，而作为案件核心的CH型生物试剂不翼而飞，此案成了一桩悬案。而就在二十多年后的现在，当年的缉毒支队成员高平率领着楚江河的儿子楚白以及一众干警势要查清当年的真相，化工广场大爆炸、四名被绑架的孩子、同一时间互相杀人的死者、摩天轮焚尸等一系列奇案的发生让真相逐渐浮出水面，但随着接近真相，一直藏在暗中的那双手使这桩跨越二十余年的大案更加扑朔迷离起来，随着刑警米乐儿的失踪，众人这才意识到，藏在黑暗背后的那双手，竟然是“他”！最终，这个尘封20年的大案终于侦破，而背后的真正元凶却是谁也未曾猜想到的。可无论如何，楚白完成了自己的梦想，为当年卧底的父亲正名，让父亲住进了烈士陵园，而自己，最终继承了父亲的警号。
